ABSTRACT

This study evaluated anesthetic efficacy and possible effects of the essential oils (EOs) of Cunila galioides (EOC) and Origanum majorana (EOO) on ventilatory rate (VR) and ionoregulation in Rhamdia quelen. In the anesthesia assessments, 50, 100, 200 and 300 μL L-1 EOC and 50, 100, 200, 300, 400 and 500 μL L-1 EOO were tested, and time for induction to sedation and anesthesia stages, as well as recovery, were taken. A second trial employed lower concentrations of both EOs, 10, 25, 50 and 100 μL L-1, in order to verify VR and Na+, K+ and Cl- whole body net fluxes. Sedation was achieved with both oils at 100 µL L-1, and anesthesia at ≥ 200 µL L-1. There was no significant difference between control and EO-treated groups regarding VR, but all fish subjected to 100 µL L-1 EOC died within 2 h of exposure. Overall, ionic loss declined in the presence of the EOs. The EOC at 200 - 300 μL L-1 and EOO at 400 - 500 μL L-1 present the potential to promote fast anesthesia in R. quelen.(AU)


No presente estudo foi avaliada a eficácia da anestesia e possíveis efeitos dos óleos essenciais (EOs) de Cunila galioides (EOC) e Origanum majorana (EOO) sobre a taxa ventilatória (VR) e regulação iônica em Rhamdia quelen. Nas avaliações de anestesia, as concentrações de 50, 100, 200 e 300 μL L-1 EOC e 50, 100, 200, 300, 400 e 500 μL L-1 EOO foram testadas, e os tempos para a indução às fases de sedação e de anestesia, assim como recuperação, foram mensurados. Um segundo ensaio empregou concentrações mais baixas de ambos EOs: 10, 25, 50 e 100 μL L-1 a fim de verificar a VR e o fluxo líquido corporal dos íons Na+, K+ and Cl- . A sedação foi alcançada para ambos os óleos em 100 μL L-1, e a anestesia em concentrações ≥ 200 µL L-1. Não houve diferença significativa entre o controle e grupos tratados com EOs em relação a VR, mas todos os peixes submetidos a 100 µL L-1 do EOC morreram dentro de 2 h de exposição. No geral, a perda iônica declinou na presença dos EOs. O EOC em 200 - 300 μL L-1 e o EOO em 400 - 500 μL L-1 apresentam potencial para anestesia rápida em R. quelen.(AU)

ABSTRACT

Adjuvants are essential to boost the immune response to inoculated antigen and play a central role in vaccine development. In this study, we investigated the efficacy of several adjuvants in the production of anti-bovine serum albumin (BSA) antibodies in silver catfish. Two hundred and seventy juvenile silver catfish (60–80 g) of both sexes were intraperitoneally vaccinated with BSA (200 µg/fish) alone or mixed to the following adjuvants: Freund’s complete adjuvant (FCA), Freund’s incomplete adjuvant (FIA), aluminum hydroxide (AlOH), Montanide, four types of cytosine-phosphate-guanine (CpG) oligodeoxynucleotides (ODNs) and three concentrations of β-glucan, and the immune enhancing property was evaluated by measuring anti-BSA antibodies in blood samples at biweekly intervals. Our results demonstrated that CpGs ODNs and β-glucan were as effective as classical adjuvants (FCA, FIA, AlOH and Montanide) in promoting anti-BSA antibodies and that the kinetics of antibody production induced by all adjuvants used in our study had a similar trend to that observed in other fish species, with a peak at 28 days post-vaccination. These results may be useful for the selection of adjuvants for vaccine formulation intended for silver catfish and for the development of vaccine and vaccination strategies to other fish species.

ABSTRACT

Light microscopic studies of the immune response of Mystus gulio were carried out. Antigen binding cells have been detected by plaque-forming cell (PFC) assay test in the spleen, head-kidney (HK) and thymus. Among these three organs, the HK is more pronounced in its response, and it is compared, on the basis of its histology of higher vertebrates. The peak response after primary and secondary immunizations was on day 7, in both circulating blood and the immune organs. The results suggest that HK in these fish might be the major organ for antibody secreting cells. The HA (haemoagglutinin) response was also for longer duration but only slightly more intense. The PFC response after the secondary immunization was for much longer duration and much more intense than after the primary immunization.

ABSTRACT

Um fator limitante nas primeiras fases do cultivo do jundiá (Rhamdia quelen Quoy e Gaimard, 1824) é a presença da ictioftiríase. Os ácidos graxos poliinsaturados (PUFAs) presentes na dieta possuem uma potente atividade imuno-moduladora, e esta atividade depende da espécie analisada e da quantidade/qualidade dos PUFAs da dieta. O objetivo do presente estudo foi avaliar a resposta fisiológica de alevinos de jundiá, alimentados com diferentes fontes de ácidos graxos, quando parasitados pelo protozoário Ichthyophthirius multifiliis (Fouquet, 1876). Alevinos de jundiá (7,6±0,8g) alimentados durante sete semanas com cinco dietas contendo diferentes fontes lipídicas: óleos de peixe, linhaça e milho foram expostos ao protozoário. No quinto dia a presença da doença foi detectada. Amostras de sangue foram coletadas e sobrevivência e graus de infestação foram estimados. As diferentes dietas afetaram a sobrevivência dos alevinos, sendo que alevinos que consumiram dietas com óleo de peixe apresentaram uma maior sobrevivência. O hematócrito e o grau de infestação não apresentaram diferença estatística significativa entre os tratamentos, entretanto a contagem diferencial dos leucócitos registrou diferença. Os resultados mostraram pela primeira vez no jundiá que, diferentes fontes lipídicas produzem respostas fisiológicas diversas, podendo representar uma alternativa ao tratamento da doença. Novos estudos deverão ser realizados no sentido de aprofundar o conhecimento aqui gerado.


A limiting factor in the early stage of the jundiá (Rhamdia quelen Quoy; Gaimard, 1824) culture is the presence of diseases such as ichthyophthiriasis or "Ich". Dietary polyunsaturated fatty acids (PUFAs) have a potent immunomodulatory activity; and this activity depend of the fish species, quantity/quality of the dietary PUFAS. The aim of this study was evaluate the physiologic response ofjundiá, rather fed diets with different sources of fatty acids when challenge to Ich disease. Fingerlings ofjundiá (7.6±0.8g) were fed during seven weeks with five diets with a different lipid source: fish, linseed and corn oils. Then, the fish were challenged with the Ichthyophthirius multifiliis (Fouquet, 1876). On the 5th day, the presence of the disease was detected. Survival rate, blood samples and infection grade were recorded. Diets affect the survival rate of fingerlings, thus fish fed with diet with fish oil showed a higher survival. Hematocrit and infestation grade no recorder differences among treatment. The differential analysis of white blood cells shows difference. The results point out, at first time injundiá, that dietary lipids caused diversity physiologic response, this may be represent an opportunity for the disease treatment. New studies must be carrying out to deep knowledge found.

ABSTRACT

Light microscopic and ultra-structural studies of the lymphoid tissues such as blood immunocytes, spleen and pronephros of the freshwater catfish, Clarias batrachus, were carried out. The peripheral blood showed nucleated erythrocytes, total leucocytic count (TLC) more than that observed in mammalian blood and leucocytes with morphological appearance similar to the mammalian white blood cells (WBCs). The spleen and pronephros showed presence of numerous lymphocytes, monocytes and nucleated red blood cells (RBCs) along with hemosiderin-containing macrophages. The morphology of lymphoid organs of the catfish has been discussed in light of the evolution of the immune system in this class of vertebrates.
